Hi Yuxin,
I am not sure what you mean by one choice for the NBS. You can either select NBS or FDR. The SPC method is not currently implemented.
Exchange blocks are used to constrain permutations across repeated measurements. They are not needed for between-group comparisons across two distinct groups. So they will not be needed for your design.
