Hi Dr. Zalesky,
eventually I've decided to use the design matrix below (2nd column treatment, 3rd condition, 4th interaction)

4-D_B 1 -1 -1 1
as you suggested I've used a t-test and separately used a contrast of 1 and -1, however I'm not sure about the meaning of the different contrasts:
0 -1 0 0 means that group drug > group placebo ?
0 0 -1 0 means that cond B > cond A ?
0 0 0 -1 what would be here the meaning? How can I define this interaction?
