Hi Lee,
You can simply ignore the 28th column (in this case the 28th ROI is likely the gray-matter-voxels ROI created by default by the conn toolbox). In general, the ROI to ROI single subject matrices will be of size m x n (with n>=m, where m is the number of 'source' ROIs -only those ROIs that have been selected as 'sources' in the 'first-level analyses' step-, and m is the total number of ROIs in your project -all ROIs except those included in the 'preprocessing' step as potential confounds-), and these matrices are sorted so that the first m columns correspond to the same ROIs as those in the first m rows.
